  • What is Desyrel?

Desyrel contain Trazodoneas active ingredient which is an oral antidepressant drug that acts to influence some chemical messengers called neurotransmitters in the brain. The job of these chemicals is to provide medium for communication (stimulate) among nerves. Depression is believed to be an outcome of imbalance of these brain chemicals. Acetylcholine, norepinephrine, dopamine and serotonin are the major neurotransmitters. However, the correct mechanism of action of Trazodone is not known, it probably recovers signs of this mental disorder by blocking the approval of serotonin by nerves. As a result of this adds serotonin to rouse other nerves. Trazodone is chemically dissimilar to the selective serotonin reuptake inhibitors (SSRIs or tricyclic antidepressants (TCAs) or to the monoamine oxidase inhibitors (MAO inhibitors). Drug is chemically similar to nefazodone (Serzone) and shares its actions. The FDA gave approval to Trazodone in 1982.

Trazodone is used for management of disorders associated with depression. Other than this, drug is sometimes also indicated in combination with other drugs for the treatment of panic attacks, aggressive behavior and cocaine withdrawal.

  • What is the generally used dose of Desyrel treatment?

Drug should be used under medical guidance as wrong use can result in serious side effect. Generally, for treatment of depression, Desyrel dose range from 150-600mg per day in adult patients. Dose is started with 150 mg per day. Trazodone is indicated for one or more daily doses, preferably before bed time. Similar to other antidepressants, drug may take several weeks for showing visible improvements in patient.

  • What are the precautions for reducing risks with the treatment of Desyrel?

In following medical conditions the use of Desyrel is not permitted or should be used with extreme caution if patient have:

  • liver or kidney disease;
  • heart disease;
  • abnormal bleeding or disorder of blood clotting;
  • seizures or epilepsy;
  • any other mental disorder (manic depression or bipolor disorder);
  • long QT syndrome;
  • drug abuse history or suicidal thoughts;
  • current history of heart attack;
  • FDA have put this drug in pregnancy category C, or
  • breast feeding a baby.

 What are the possible drug interactions of Desyrel with other drugs that one should be careful of while taking treatment?

  • other antidepressant medicines;
  • bosentan;
  • imatinib;
  • isoniazid;
  • St. John's wort;
  • methadone;
  • tacrolimus;
  • arsenic trioxide;
  • antibiotic drugs such as azithromycin, erythromycin, clarithromycin,levofloxacin, pentamidine, moxifloxacin;
  • antifungal medication--itraconazole, ketoconazole;
  • anti-malaria medication
  • blood thinner including warfarin, Coumadin;
  • heart rhythm medicine
  • hepatitis C medications--boceprevir, telaprevir;
  • HIV/AIDS medication--atazanavir, efavirenz, indinavir, ritonavir, saquinavir;
  • Drug to manage psychiatric disorders such as chlorpromazine, mesoridazine, pimozide, thioridazine;
  • migraine headache medicine;
  • seizure medication such as carbamazepine, oxcarbazepine, phenytoin, primidone; or
  • tuberculosis medication such as rifabutin, rifampin, rifapentine.

 What are the possible side effects of Desyrel treatment?

Common side effects associated with Desyrel (>10%)

  • Blurred vision (11-15%)
  • Dizziness (20-28%)
  • Drowsiness (24-40%)
  • Dry mouth (15-34%)
  • Fatigue (11-15%)
  • Headache (16-20%)
  • Nausea/vomiting (11-15%)

Less common unwanted side effects of Desyrel (1-10%)

  • Constipation
  • Edema
  • Confusion
  • Disorientation
  • Incoordination
  • Nasal congestion
  • Orthostatic hypotension
  • Syncope
  • Tremor
  • Weight change

Some other critical side effects (occurrence not known)

  • Priapism
  • Sedation

 Is Desyrel safe to be used in children?

Desyrelis not approved to be used in pediatric patients. Short-term studies have shown that antidepressant drugs raise the risk of suicidal thinking and behavior in pediatric patients, and young adults (<24 year of age) using this antidepressants for depressive disorders.

  • What are the contents of Desyrel?

Tablet of DESYREL (trazodone hydrochloride) contains microcrystalline cellulose, FD&C Yellow No. 6 (aluminum lake), magnesium stearate, pregelatinized starch, and stearic acid as inactive ingredient.
