MECHANISM OF ACTION

Sildenafil is used to treat erectile dysfunction. cGMP is a chemical in the body which causes relaxation of vascular smooth muscles and increases blood flow to various tissues of the body.  PDE5 (phosphodiesterase5) is an enzyme which carries out the breakdown of cGMP. It is found in the erectile tissue of the penis, vascular endothelium (walls of blood vessels) and retina of the eyes. Sildenafil increases the concentration of cGMP in tissues by inhibiting PDE5. Increased cGMP cause vasodilation of penile blood vessels which results in erection.
